Previously Glen Research noted the use of lipid-conjugates as lipophilic carriers of oligonucleotides and added 5'-stearyl phosphoramidite to their amidite portfolio for modifying small interfering RNA (siRNA) and other oligonucleotides.

Researchers have demonstrated the utility of such hydrophobic and lipid-modified oligonucleotides, and more recently have shown that lipid-oligo conjugates with more than one fatty chain modification can enhance the delivery of siRNA in vivo, and in some cases facilitate tissue-specific delivery.

This white paper from Glen Research briefly summarizes some of the important advances in lipid carriers of RNA molecules and describes a recent study of lipid valency, the number of fatty acid chains attached to an siRNA molecule, and its impact on tissue-specific delivery and gene expression.
