Regulatory (Treg) cells are a critical subpopulation of CD4 T cells that suppress immune response and maintain peripheral homeostasis and immunological tolerance. Imbalanced Treg frequency and dysregulation of Treg function may lead to the development of chronic inflammatory diseases and cancer. Knowing the crucial role of Tregs in immune tolerance, Treg modulation has become an important therapeutic approach to treating immune disorders and preventing transplantation rejection in clinical settings. Despite increased numbers of studies on the function of Tregs, their heterogeneity and mechanism of differentiation remain unclear.
